If i have a demand of 400 on a route, is it better to have 2 X 737 or 1 X 757 doing the route?

No but does it matter and why?

Frequency always wins.  If you have 4 737s and someone else has 2 757s, you'll get the majority of the pax.  And vice versa.

Pax love choice!

But if im the only one on that route??

frequency = win

if he fly 2x 737 and you 1x 757 then he will get 66% of pax.

If there is no competition, yes, fewer flights with bigger aircraft would save money (expenses).  But if there is competition, and you lose passengers to competition flying with higher frequency, you will lose a lot more revenue than you save in expenses.

My point exactly for asking.... I would probably stick with the 737s b/c you may indeed have competition and this way you will already have the upper hand.
