Is it possible to assign different aircraft to same route
for example flight is everyday, single route #, but assign 7 different ac to the route?
Yes, just create a one-day route, create it for the following six days and then assign every single day to a desired aircraft.
cammellodacorsa, doing that would create 7 different flight numbers. I'm wondering if one flight # can utilize different ac
Below is route info for LBG-JFK.
Owl Express flies a DC 10 , same route, same plane, same flight # 6 days a week.
My DC10 cannot so this. Actually flight time round trip is more than 24 hours plus turn time
How is Owl Express doing this?

open 7 times a tab with "open new route" on the same line - one per da of the week. Create your 7 lines. If they go the same direction, then they'll have the same number.
Make sure they don't start the same day.
Works even for flights at different times of the day. I just made it in GW3 on the Faro-Brasilia route. I've got 7 different start times, yet the same line number. Though like that it appears with 7 lines on the line screen. If they were at the same time, it would be just 1 line.

do this first, and once you've done it, how to make the same at the same time is obvious.
You can assign the same flight number to a daily flight to a particular destination when its broken up into individual flights. Here's what my scheduling page looks like for the route that jvratsinas posted about of mine. I would have 100% the same flight numbers, but they auto dialed up and scrambled themselves when i was scheduling and i didn't care to fix it.
