Your initial slots are either free or very cheap.  Subsequent slots get more and more expensive dependent on a number of variables including time as well as whether you are monopolizing the arrival/destination airport or route, but these costs level out very quickly and they are a tiny portion of the potential profits on a route regardless, so they're essentially a moot point.

I have also seen airports where no one is flying, where the slot cost has been $0  ;)
